The Il12rb1 gene encodes a type I transmembrane protein that belongs to the hematopoietin receptor superfamily. This protein binds to interleukin-12 (IL-12) with low affinity and participates in IL-12 signal transduction. When combined with IL12RB2, it forms a functional high-affinity receptor for IL-12. The co-expression of this protein and IL12RB2 has been shown to create high-affinity IL-12 binding sites and reconstruct IL-12-dependent signal transduction. Mutations in the Il12rb1 gene impair the development of T lymphocytes that produce interleukin-17 and increase susceptibility to mycobacterial and Salmonella infections. Il12rb1 knockout (KO) homozygous mice exhibit reduced serum levels of interferon-gamma (IFN-γ) in response to recombinant IL-12 or lipopolysaccharide (LPS) treatment. Additionally, splenocytes from these mice activated by concanavalin A (ConA) in response to IL-12 cannot proliferate or secrete IFN-γ[1]. These homozygous mice are viable and fertile.
