The IL7 gene encodes the cytokine Interleukin-7 (IL-7), a secreted protein crucial for the development, survival, and homeostasis of B and T lymphocytes, as well as innate lymphoid cells (ILCs) [1]. Gene expression of IL7 is primarily observed in stromal cells of primary and secondary lymphoid organs like the bone marrow and thymus, as well as in non-marrow-derived epithelial cells in various peripheral tissues, including the intestine and skin. The function of the encoded IL-7 protein is to bind to its receptor (composed of the IL-7R α-chain and the common γc-chain) on lymphoid progenitor and mature cells, activating the JAK-STAT signaling pathway to promote cell proliferation and survival [2]. Dysregulation of the IL-7/IL-7R pathway is linked to several associated diseases, most notably severe combined immunodeficiency (SCID) due to T-cell deficiency, as well as various autoimmune diseases (e.g., Multiple Sclerosis, Rheumatoid Arthritis, Type 1 Diabetes) and lymphoid malignancies like Acute Lymphoblastic Leukemia (ALL) [3].
B6-hIL7 mouse is a humanized model generated using gene editing technology, in which the coding sequence of exon 1 is replaced with the Kozak-Human IL7 CDS-3'UTR of Mouse Il7-WPRE-BGH pA cassette. This model can be used for studying the pathological mechanisms and therapeutic approaches of severe combined immunodeficiency (SCID), various autoimmune diseases, and lymphoid malignancies, as well as for the development of IL7-targeted drugs.
