품종 계통 설명
The Cadherin 16 (CDH16) gene encodes a calcium-dependent, membrane-associated glycoprotein that belongs to the calmodulin superfamily. This cell adhesion molecule is involved in embryonic development and cell growth and is exclusively expressed in the kidney, where it is also known as kidney-specific calmodulin (ksp-calmodulin). CDH16 functions as a major mediator of homotypic cell recognition and plays a role in directing tissue morphogenesis, particularly in the formation of renal tubules during embryonic development.
Cdh16-MerCreMer mice were generated by inserting a tamoxifen-inducible Cre recombinase protein (MerCreMer) expression element downstream of the stop codon of the mouse Cdh16 gene. The expression pattern of MerCreMer mirrored that of the endogenous gene. Before induction, MerCreMer was localized to the cytoplasm, and only after tamoxifen treatment could it translocate to the nucleus to exert its recombination effect. When bred with mice containing a loxP site-flanked sequence, Cre recombinase-mediated deletion of the flanked sequence will occur in the renal tubular epithelial cells of the offspring following tamoxifen induction.
변형 전략
The "IRES-MerCreMer" gene expression element was inserted into the 3'UTR of the mouse Cdh16 gene using gene editing technology.

1. Method
Sperm from Cdh16-MerCreMer mice were used to fertilize oocytes from Rosa26-LSL-tdTomato mice, generating double-heterozygous offspring. At 6 weeks of age, these offspring were subjected to Tamoxifen induction via gavage at a dose of 4 mg per mouse for 5 days. This resulted in Cre recombinase-mediated deletion of LSL elements and subsequent expression of tdTomato in Cre-positive cells. One week after induction, lung, and renal tissues were collected from the offspring. The distribution of tdTomato protein was determined by immunofluorescence staining and immunohistochemistry to assess the expression of Cre recombinase.
2. Groups
Cre+Tam+: Cdh16-MerCreMer [KI/+]; Rosa26-LSL-tdTomato [CKI/+], with Tamoxifen;
Cre+Tam-: Cdh16-MerCreMer [KI/+]; Rosa26-LSL-tdTomato [CKI/+], without Tamoxifen.
3. Result
(1)Immunohistochemistry of renal tissue
The presence of tdTomato-positive signals in renal tubular epithelial cells of tamoxifen-induced Cdh16-MerCreMer[KI/+];Rosa26-LSL-tdTomato[CKI/+] mice indicates strong expression of Cre recombinase in renal tissues. In contrast, no tdTomato expression or Cre recombinase activity was observed in these tissues in the non-tamoxifen-induced group.

Figure 2. Results of immunohistochemistry of renal tissue.
(2)Immunofluorescence staining of renal tissue
The presence of a large amount of tdTomato-positive signal in the renal tubular epithelial cells of tamoxifen-induced Cdh16-MerCreMer [KI/+];Rosa26-LSL-tdTomato[CKI/+] mice indicates strong Cre recombinase activity at this site. However, a trace amount of tdTomato expression was also observed in the uninduced group, suggesting that this model exhibits a small amount of production leakage in renal tubular epithelial cells.

(3)Immunofluorescence staining of lung tissue
The presence of tdTomato-positive signals in some lung cells of tamoxifen-induced Cdh16-MerCreMer[KI/+];Rosa26-LSL-tdTomato[CKI/+] mice indicates the presence of a small amount of Cre recombinase activity at this site. In contrast, no tdTomato-positive signals or Cre recombinase expression were observed in the uninduced group.

4. Summary
In Cdh16-MerCreMer mice, Cre recombinase expression is primarily localized to renal tubular epithelial cells, with some activity also observed in lung tissue cells. It should be noted that a small amount of Cre recombinase expression leakage occurs in renal tubular epithelial cells before induction. This model represents a highly tamoxifen-inducible, tissue-specific Cre mouse line targeting renal tubular epithelial cells.
