The EGFP protein in EGFP Reporter mice is systemically expressed under the regulation of the CAG promoter, an artificially constructed combinatorial promoter consisting of the cytomegalovirus (CMV) early enhancer element and the chicken beta-actin promoter, which is commonly used to drive high-level expression of genes in expression vectors in mammals [1]. This strain allows for the direct observation of green fluorescence emitted by the EGFP protein in the presence of excitation light. Previous studies have shown that EGFP protein is not expressed in the hair and blood of multiple EGFP reporter models [2-3]. However, our results showed that the EGFP protein was expressed at a high level in multiple blood cells of this strain, suggesting that the EGFP protein in this strain has a broader expression range.

Transgene (TG) targeting technology was used to insert the EGFP gene expression sequence into the pCAGGS vector, which consists of the chicken β-actin promoter, the CMV early enhancer, the β-actin intron, and the bovine globulin polyA signal.

1. Systemic expression of EGFP in neonatal mice
Systemic green fluorescence expression in this mouse can be observed under excitation light irradiation alone.

2. Expression of EGFP in various tissues
Tissues of the heart, liver, spleen, lung, kidney, muscle, testis, intestine, fat, and other tissues of EGFP mice were taken, and the expression of green fluorescence was observed under the fluorescence microscope. The results showed the presence of green fluorescence in almost all tissues of EGFP Reporter mice.

3. Expression of EGFP in the hair
The results showed that, similar to previous studies, there was no expression of EGFP in the hair of EGFP Reporter mice.

4. Expression of EGFP protein in blood cells
Expression of EGFP protein was detected in T cells (CD45+CD3+), CD4+ T cells (CD45+CD3+CD4+), CD8+ T cells (CD45+CD3+CD8+), B cells (CD45+CD19+), and myeloid cells (CD45+CD11b+) in peripheral blood, thymus, and spleen by flow cytometry. The results showed that strong EGFP expression was detected in T cells and B cells in peripheral blood, thymus, and spleen, with over 50% of B cells positive for EGFP and an average positive rate of over 30% for T cells. In addition, over 70% of thymic CD4+ and CD8+ T cells expressed EGFP protein. Myeloid cells in peripheral blood and spleen also maintained a high level of EGFP positivity.
