The Pvalb gene encodes the protein parvalbumin, a high-affinity calcium-binding protein structurally and functionally similar to calmodulin and troponin C, and is thought to be involved in muscle relaxation. Parvalbumin is primarily expressed in the kidneys and brain, and its expression in the nervous system is fundamental. It participates in excitatory and inhibitory chemical synaptic transmission and maintains normal neural function.
Pvalb-T2A-iCre mice were generated using gene editing technology to integrate a T2A-iCre expression cassette into the stop codon of the mouse endogenous Pvalb gene. Under the control of the Pvalb gene's regulatory elements, the iCre recombinase (a codon-optimized Cre recombinase) is expressed. The expression pattern of iCre recombinase is similar to that of the endogenous Pvalb gene. Using the 2A peptide for linkage avoids the drawbacks of low protein activity and low downstream gene expression in multi-gene expression processes. When Pvalb-T2A-iCre mice are crossed with mice containing loxP sites, Cre recombinase-mediated recombination between loxP sites will occur in the neurons expressing parvalbumin protein in the offspring.

1. Method
Pvalb-T2A-iCre mice were crossed with Rosa26-LSL-tdTomato mice, which conditionally express the tdTomato fluorescent protein, to generate double-heterozygous offspring. Deletion of the stop element (LSL) mediated by Cre recombinase resulted in tdTomato protein expression in Cre-positive cells. When offspring mice reached 6 weeks, tissues from the brain, ovaries, and testes were collected. The expression of the tdTomato protein was examined using immunofluorescence staining (IF) to determine the expression of the Cre recombinase.

3. Result
(1)Expression of Cre recombinase in the cerebellum and olfactory bulb
Histological morphological analysis shows a substantial green fluorescent signal in the Purkinje cells of the cerebellum in Cre+ mice, indicating that Cre recombination has occurred in these cells. Additionally, a significant green fluorescence is observed in the EPL and GCL regions of the olfactory bulb in Cre+ mice, indicating Cre recombination in these cells.

(2)Expression of Cre recombinase in the cortex and hippocampus
Histological morphological analysis shows a substantial green fluorescent signal in the cortex and hippocampus of Cre+ mice, indicating that Cre recombination has occurred in these tissues. A significant green background is observed in the experimental group.

(3)Expression of Cre recombinase in the testis and ovary
The results show a substantial green fluorescence in the germ cells of the testes of Cre+ mice, indicating Cre recombination in these cells. Additionally, internal PCR results indicate the presence of a distinct CreDel band in the epididymis and testes of Cre+ mice (not shown in this manual). Combining the PCR and IF results suggest the possibility of male germline leakage in Pvalb-T2A-iCre mice. No green fluorescence was observed in the ovarian tissues of Cre+ mice, indicating that Cre recombination did not occur in these tissues.

4. Summary
In the Pvalb-T2A-iCre mouse model, Cre recombinase is significantly expressed in the brain tissues of adult mice (cerebellum, olfactory bulb, cortex, hippocampus). Notably, there may also be male germline leakage in this mouse model, where Cre recombination might occur in the male germ cells.
