The CRBN gene, located on human chromosome 3, exhibits broad expression across diverse tissues, including the brain, kidney, muscle, and immune cell populations such as monocytes, macrophages, dendritic cells, and B lymphocytes [1]. This gene encodes cereblon, a protein that functions as a key substrate receptor within the CRL4-CRBN E3 ubiquitin ligase complex. This complex mediates the ubiquitination and subsequent proteasomal degradation of specific target proteins, thereby regulating crucial cellular processes encompassing protein homeostasis, ion transport, and AMPK signaling [1-2]. Notably, mutations in CRBN are implicated in autosomal recessive nonsyndromic intellectual disability [2]. Furthermore, Cereblon protein serves as a primary molecular target for targeted protein degradation (TBD) therapy by specifically modulating the enzymatic activity of the CRL4-CRBN complex and altering its substrate recognition properties, thereby enabling the selective degradation of specific transcription factors. This molecular mechanism has emerged as a critical theoretical foundation for the clinical treatment of malignant hematological diseases, such as multiple myeloma, leading to the development of diverse therapeutic modalities including molecular glues and proteolysis targeting chimeras (PROTACs) [3-5].
hCRBN mice are humanized models generated by gene editing technology, in which the exon 2 to partial intron 2 of the mouse Crbn gene was replaced in situ with the Exon 2~11 of the coding sequence (CDS) of human CRBN gene. This model can be used to study the pathological mechanisms and therapeutic methods of autosomal recessive nonsyndromic intellectual disability (ARNSID), multiple myeloma and other hematological cancers, as well as the screening, development, and preclinical efficacy and safety evaluation of CRBN-based targeted protein degradation (TBD) therapies.

1. Gene Expression
Thymus and brain tissues were collected from homozygous hCRBN and wild-type (WT) mice. Human CRBN and mouse Crbn transcripts were quantified using species-specific primers, with mGapdh as the reference gene. Relative expression levels were calculated and presented as mean±SD. RT-qPCR results confirmed the exclusive detection of human CRBN transcripts in homozygous hCRBN mice, whereas only mouse Crbn transcripts were detected in WT mice.

2. Protein Expression
Western Blot (WB) results showed that human CRBN protein was detected in the thymus, spleen, and brain of homozygous hCRBN mice, with decreased expression observed in the brain tissue.

*A cross-reactive antibody recognizing both murine and human CRBN proteins was used in this experiment.
3. CC-885 Toxicity Assessment and GSPT1 Degradation Assay
(1)CC-885 Toxicity Assessment
CC-885 is a CRBN molecular glue compound that exhibits potent antitumor activity by inducing the degradation of essential proteins, such as GSPT1. To evaluate the toxicity of the CRBN modulator CC-885 in hCRBN mice, wild-type (WT) and homozygous hCRBN mice were divided into two groups and treated intraperitoneally with 5 mg/kg CC-885 or vehicle control for seven consecutive days. (A) Percentage of mouse survival; (B) Body weight during the experimental period.
Results showed that all CC-885-treated hCRBN mice died within 50 hours post-injection, whereas all CC-885-treated WT mice and vehicle controls survived. These findings indicate that CC-885 exhibits significant toxicity exclusively in hCRBN mice, with no overt toxicity observed in WT mice.

(2)GSPT1 Degradation Assay
9-week-old female wild-type (WT) and homozygous hCRBN mice were divided into two groups and treated intraperitoneally with 5 mg/kg CC-885 or vehicle control for seven consecutive days. Spleen tissues were collected at the experimental endpoint to prepare tissue lysates. Western blot analysis was performed using antibodies against CRBN, GSPT1, CK1ε, and GAPDH (loading control). CRBN was used to validate the humanized model; GSPT1 to confirm target degradation; and CK1ε as a reference negative control (two bands observed at 40–55 kDa with consistent trends).
Western blot results showed no significant changes in CK1ε expression across treatment groups, while CRBN levels decreased slightly in the CC-885-treated hCRBN group (G4). GSPT1 protein was detected in the vehicle-treated WT (G1) and hCRBN (G3) groups, as well as in the CC-885-treated WT group (G2), but was undetectable in the CC-885-treated hCRBN group (G4). These results indicate that CC-885 induces GSPT1 degradation in the spleen of hCRBN mice, an effect not observed in WT mice.
