Bex2, or Brain Expressed X-linked gene 2, was originally identified for promoting transcription by interacting with transcription factors in DNA-binding complexes. It is involved in multiple cellular processes and associated with several signaling pathways such as NF-κB, c-Jun/JNK, and ErbB2 pathways [2]. Bex2 has been shown to play a significant role in carcinogenesis, making it biologically important in understanding cancer development [2]. Genetic models like gene knockout (KO) could potentially be valuable in further elucidating its functions.
Knockdown of Bex2 in various cancer cell lines has revealed its role in cancer-related processes. In lung cancer cells, crotonylated Bex2 enhances mitophagy by facilitating the interaction between NDP52 and LC3B, and its overexpression inhibits chemotherapy-induced apoptosis [1]. In breast, colorectal, hepatocellular cancers, and brain tumors, Bex2 promotes cell growth, survival, migration, and invasion, and regulates apoptosis and the cell cycle through cross-talk with relevant signaling pathways [2]. In gastric and hepatocellular carcinomas, Bex2 is associated with cancer stemness and poor prognosis, and its knockdown decreases stem-like properties and increases drug sensitivity [3,4]. In colorectal cancer, Bex2 promotes cell proliferation via the JNK/c-Jun pathway [5]. In intrahepatic cholangiocarcinoma, Bex2 is essential for maintaining dormant cancer stem cells by suppressing mitochondrial activity [6]. In glioma cells, Bex2 promotes migration and invasion, likely mediated by N-cadherin and MMP-2 [7].
In conclusion, Bex2 is a gene with diverse functions in cancer biology, mainly promoting cancer-related processes such as cell growth, survival, migration, and invasion, and is associated with cancer stemness and poor prognosis in multiple cancer types. The use of knockdown and overexpression models in various cancer cell lines has significantly contributed to understanding Bex2's role in these disease conditions, providing potential therapeutic targets for cancer treatment.
