Cd82, also known as KAI1, is a glycoprotein belonging to the tetraspanin family. It functions as a tumor metastasis suppressor, inhibiting the metastasis of most tumors [1,2,4,6,7,8,10]. It is involved in multiple cellular processes such as signal transduction, cell adhesion, motility, and aggregation [5]. The loss of its expression is often associated with advanced stages of many human cancers and acquisition of metastatic potential [7].
In a Cd82 -/- mouse model, it was found that CD82 deficiency decreased the severity of colitis in mice. CD82 was shown to suppress the activation of the NLRP3 inflammasome, and its deficiency led to increased NLRP3 deubiquitination by BRCC3, reducing colitis pathogenesis [3]. In young adult mice, injection of AAV-CD82 to the hippocampus induced AD-like cognitive deficits, while hippocampus-related memory functions were improved in Cd82 -/- mice, indicating that elevated CD82-TRPM7-Numb signaling is linked to age-related cognitive impairment [9].
In conclusion, Cd82 plays a crucial role as a tumor metastasis suppressor, influencing cancer progression. Mouse models, such as Cd82 -/- mice, have revealed its functions in colitis and age-related cognitive impairment, contributing to our understanding of these disease areas and potentially paving the way for targeted therapies [3,9].
