TARM1, short for T cell-interacting, activating receptor on myeloid cells 1, is a member of the leukocyte immunoglobulin-like receptor family. It associates with FcRγ and is involved in stimulating macrophages and neutrophils. TARM1 participates in pathways related to inflammation, such as NF-κB pathway, and plays a role in immune-related biological processes [3].
In collagen-induced arthritis (CIA) mouse models, Tarm1-/-mice show suppressed development of CIA, with impaired T cell priming against type 2 collagen and antigen-presenting ability of GM-CSF-induced dendritic cells, indicating TARM1's role in promoting arthritis development [1].
In a mouse colitis model, knockdown of Tarm1 in RAW264.7 cells inhibits lipopolysaccharide (LPS)-induced M1 polarization of macrophages and attenuates intestinal barrier damage [2].
In Mycobacterium tuberculosis infection, TARM-1 knockdown in macrophages reduces pro-inflammatory cytokine expression, HLA-DR and costimulatory molecules, and TARM-1 blocking peptide in mouse models increases bacterial load and lung pathology, suggesting its importance in macrophage activation and Th1 response in this infection [4].
In conclusion, TARM1 is an important factor in the immune system, playing roles in diseases like arthritis, colitis, and tuberculosis. Gene knockout models in mice have been crucial in revealing its functions in these disease-related biological processes, highlighting its potential as a therapeutic target for autoimmune and infectious diseases.
