Rbm47, an RNA-binding protein, contains three RNA recognition motif (RRM) domains. It is essential for embryonic endoderm development in vertebrates [3,4]. Rbm47 is involved in multiple biological processes such as RNA editing, immune homeostasis, and is associated with pathways like Wnt/β-catenin, FGF-ERK, and antiviral immune response pathways [3,4,5]. Genetic models, especially KO/CKO mouse models, have been crucial for studying its functions.
Intestine-specific Rbm47-knockout (Rbm47-IKO) mice showed increased proliferation, abnormal villus morphology, and cellularity. They adapted to radiation injury, were protected against chemical-induced colitis and colitis-associated cancer, but aged Rbm47-IKO mice developed spontaneous polyposis, and Rbm47-IKO ApcMin/+ mice had an increased intestinal polyp burden. This indicates Rbm47 modifies intestinal proliferation, oxidative response, and inflammatory pathways [1]. K329R knockin (KI) mice with defective RBM47-ISGylation displayed heightened susceptibility to LPS-induced acute lung injury and lung tumorigenesis, accompanied by immunosuppression, suggesting that RBM47-ISGylation is important for immune activation and lung homeostasis [2]. Down-regulation of Rbm47 in mouse embryonic stem cells (mESCs) perturbed the expression of primitive endoderm (PrE) markers, increased GATA4 + PrE-like cells, and affected multi-lineage differentiation potential, showing its role in mESC differentiation [4].
In conclusion, Rbm47 is essential for embryonic development, cell-fate decisions in mESCs, and maintaining immune homeostasis. The KO/CKO mouse models have revealed its role in various disease conditions such as intestinal injury, tumorigenesis, lung injury, and cancer, providing valuable insights into the underlying mechanisms and potential therapeutic targets for these diseases.
