Mxra7, Matrix Remodeling Associated 7, is a gene related to matrix remodeling processes. It is involved in various biological functions, potentially through associated signaling pathways such as ERK/MAPK, NF-κB, and others. Its study is crucial as it impacts multiple physiological and pathological processes [1,2,3].
In gene knockout studies, Mxra7-/-mice showed reduced megakaryocytes in the bone marrow and spleen, decreased platelet count and function in peripheral blood. Knock-out inhibited the differentiation of hematopoietic stem/progenitor cells to megakaryocytes by down-regulating GATA-1 and FOG-1 [1]. In acute promyelocytic leukemia cells (NB4), knockdown of Mxra7 promoted drug-induced apoptosis and ATRA-induced cell differentiation, while overexpression had opposite effects. In B-ALL cell line REH, knockdown inhibited cell proliferation and increased sensitivity to cytarabine [3,4]. In SHI-1 cells, overexpression of Mxra7 reduced drug-induced apoptosis by increasing BCL-2 expression [5]. In CCl4-induced acute liver injury in mice, Mxra7 deficiency alleviated liver damage, while overexpression aggravated it. Mxra7-/-mice also showed reduced neutrophil recruitment, CD8+ T-cell number, and cytokine release, along with suppressed MAPK and AKT/NF-κB pathways [6]. In cutaneous wound healing, MXRA7-deficient mice had impaired wound healing, with reduced fibroblast migration, contraction, and proliferation [7]. In psoriatic epidermis, MXRA7-deficient mice developed severer psoriasis-like diseases [8]. In bone marrow mesenchymal stem cells, MXRA7-deficient mice had retarded osteogenesis, and cultured MXRA7-deficient BMSCs showed decreased osteogenesis upon induction [9].
In conclusion, Mxra7 plays essential roles in processes like megakaryocyte differentiation, platelet production, cell apoptosis and differentiation in leukemia, liver injury, wound healing, psoriasis, and bone formation. Gene knockout mouse models have been instrumental in revealing its functions in these disease-related areas, providing potential new targets for treating platelet-related diseases, leukemia, liver injury, psoriasis, and bone-related disorders [1,3,4,5,6,7,8,9].
