Prxl2a, also known as peroxiredoxin like 2A, is an antioxidant protein. It protects cells from oxidative stress and may be involved in multiple biological processes related to redox regulation [1]. Oxidative stress is associated with various disease conditions, highlighting the importance of Prxl2a in maintaining cellular homeostasis.
In oral squamous cell carcinoma (OSCC), miR-125b downregulation leads to Prxl2a upregulation, protecting tumor cells from oxidative stress. Exogenous miR-125b expression in OSCC cells increases oxidative stress and drug sensitivity, similar to Prxl2a knockout, indicating an miR-125b-Prxl2a regulatory axis in OSCC pathogenesis [1].
In endometrial cancer, a prognostic signature including Prxl2a was established. Knockdown of Prxl2a significantly inhibited the proliferation and colony formation of Ishikawa and AN3CA cells, and suppressed xenograft growth [2].
In kidney renal clear cell carcinoma, Prxl2a was identified as one of six antioxidant genes significantly correlated with patient outcome, and high-risk patients (related to the six-gene signature including Prxl2a) had poorer prognosis [3].
In an Alzheimer's mouse model treated with cyanidin 3-O-glucoside, Prxl2a was among the upregulated antioxidant genes, suggesting its role in the antioxidant mechanism of the treatment [4].
In a mouse model of snakebite envenoming, Prxl2a was predicted as a biomarker sensitive to antivenom treatment, indicating its potential role in the pathophysiological changes after envenoming [5].
In conclusion, Prxl2a plays a crucial role in protecting cells from oxidative stress. Through gene-knockout or knockdown models in different disease conditions such as OSCC, endometrial cancer, kidney renal clear cell carcinoma, Alzheimer's disease, and snakebite envenoming, its functions in promoting tumor cell survival, affecting prognosis, and being involved in antioxidant and pathophysiological processes have been revealed. These findings contribute to a better understanding of the molecular mechanisms underlying these diseases and may provide potential therapeutic targets.
