Tgfbrap1, also known as TGF-β receptor-associated binding protein 1, plays crucial roles in multiple biological processes. It is involved in the TGF-β signaling pathway, which regulates various cellular functions such as cell growth, differentiation, apoptosis, and extracellular matrix production. Dysregulation of this pathway can lead to numerous diseases [1,5,7].
In pulmonary fibrosis, Chitinase 1 (CHIT1) enhances TGF-β1-stimulated fibrotic responses through inhibiting TGF-β1 induction of SMAD7. This effect is mediated by CHIT1's interaction with Tgfbrap1, highlighting Tgfbrap1's role in the pathogenesis of pulmonary fibrosis [1,7].
In goose follicular granulosa cells, knockdown of Tgfbrap1 mRNA level reduces apoptosis rate and enhances the secretion of E2 and P4, indicating its role in regulating follicular development [2].
Also, a variant at Tgfbrap1 (rs2241797) is significantly associated with type 2 diabetes mellitus and affects diabetes-related miRNA expression [3].
Additionally, the A variant of rs17687727 in Tgfbrap1 influences the susceptibility to antituberculosis drug-induced liver injury in the Han Chinese population [4].
In liver cancer, Tgfbrap1 stabilizes TGF-β receptor type 1 (TGFBR1), potentiating TGF-β signaling and promoting liver cancer stemness and drug resistance [5].
Piperine, which protects against acetaminophen-induced hepatotoxicity, may mediate its effects through regulating Tgfbrap1 [6].
Overexpression of Tgfbrap1, a CORVET-specific subunit, decreases the amount of assembled HOPS complex, affecting autophagosome-lysosome fusion and lysosomal hydrolase delivery [8].
