Flot1, also known as Flotillin-1, is a scaffold protein of lipid rafts. It is involved in multiple biological processes such as lipid raft protein-dependent or clathrin-independent endocytosis, and the formation of hippocampal synapses. It participates in signaling transduction and vesicular trafficking pathways, playing a significant role in normal physiological functions and disease development [2].
FLOT1 has been found to have a profound impact on cancer progression. In gastric cancer, FLOT1 promotes cell progression and metastasis by interacting with BCAR1 through the ERK signaling pathway [1]. In lung adenocarcinoma, it promotes tumor development, induces epithelial-mesenchymal transition, and modulates the cell cycle by regulating the Erk/Akt signaling pathway [5]. In acute myeloid leukemia (AML), increased FLOT1 expression is correlated with poor clinical outcome, and its knockdown inhibits AML cell growth through triggering apoptosis and pyroptosis [4]. In glioblastoma, the SDC1-TGM2-FLOT1-BHMT complex influences the fusion of autophagosomes with lysosomes, determining radiosensitivity [3]. In esophageal cancer, interfering with Circ_0003855 expression, which is related to miR-622 and FLOT1, can inhibit the activity of cancer cells [6]. In ovarian cancer, FLOT1 is a novel serum biomarker, and its mRNA m6A modification can be targeted for inhibition [7].
