Tada1, also known as Hfi1 in Cryptococcus neoformans, is involved in multiple biological processes. It is a part of the core module of the Spt-Ada-Gcn Acetyltransferase (SAGA) complex, which is a conserved co-activator in eukaryotes. Tada1 is important for maintaining the structural integrity of the SAGA complex [5].
In wheat, TaDA1 (a form of Tada1) acts as a conserved negative regulator of kernel size. Overexpression of TaDA1 decreases kernel size and weight, while its down-regulation has the opposite effect. TaDA1-A physically interacts with TaGW2-B, and they have additive effects on kernel weight [2].
In human cells, Tada1 has been associated with cancer-related processes. In a melanoma model, it was identified as a gene whose loss is involved in resistance to vemurafenib through genome-scale CRISPR-Cas9 knockout screening [1]. Also, in colorectal cancer, miR-7702 acts as a tumor suppressor by directly inhibiting TADA1, suppressing cell migration and invasion [3]. In lung squamous cell carcinoma, LINC00511 promotes tumor progression partially by up-regulating TADA1 via targeting miR-150-5p [4]. Moreover, in ovarian cancer cell lines, TADA1 was screened as a novel gene potentially involved in cisplatin resistance using a Genome-scale CRISPR-Cas9 knockout library [6].
In conclusion, Tada1 has diverse functions across different species. In plants like wheat, it regulates kernel size and weight. In humans, it is implicated in cancer-related processes such as drug resistance, cell migration, and invasion. The use of techniques like gene knockout in screening has been crucial in uncovering these roles of Tada1 in different biological contexts.
