GLG1, also known as Golgi glycoprotein 1, is an intracellular fibroblast growth factor receptor located in the Golgi apparatus [2]. It may function as a decoy FGF receptor [2]. GLG1 is associated with various biological processes and diseases. In cancer, it has been studied in relation to Ewing sarcoma, malignant glioma, breast cancer, and gastric cancer [1,2,3]. It is also involved in estrogen synthesis regulation in mouse ovarian granulosa cells [4], and a circular RNA derived from GLG1 mRNA (circ_GLG1) plays a role in colorectal cancer progression [5].
In Ewing sarcoma, high BCL11B and/or GLG1 immunoreactivity in CD99-positive cases shows high specificity (97.5%) and accuracy (87.4%) for diagnosis by immunohistochemistry, helping as specific auxiliary markers in diagnosing Ewing sarcoma especially when confirmatory molecular diagnostics are unavailable [1]. In malignant glioma and breast cancer cells, memantine treatment up-regulates GLG1 mRNA expression, changes its splicing variant profiles, and alters its cellular location, suggesting a role in the cancer-suppressive effects of memantine [2]. In gastric cancer, FUT3 promotes cell migration by synthesizing Lea on GLG1, affecting its distribution in intracellular vesicles [3]. In mouse ovarian granulosa cells, taurine down-regulates GLG1, and GLG1 knockdown increases estrogen synthesis, indicating its role in the p38/miR-7a/Glg1/Cyp19a1 signaling pathway for estrogen synthesis [4]. In colorectal cancer, circ_GLG1 promotes KRAS expression by acting as an miR-622 sponge, affecting cancer cell viability, proliferation, invasion, and migration [5].
In summary, GLG1 is involved in multiple biological processes and diseases. Its study in different cancer types and estrogen synthesis regulation provides insights into its functions. Research on GLG1, especially through the findings in these disease-related studies, helps understand the underlying molecular mechanisms, which may contribute to the development of new diagnostic and therapeutic strategies for related diseases.
