NAPG, encoding gamma-SNAP, is one of three soluble N-ethylmaleimide-sensitive fusion (NSF)-attachment proteins (SNAPs). It is located on chromosome 18p11 and is thought to play a role in cellular processes required for neurotransmission in the central nervous system [6].
In a study of a Chinese pedigree with hereditary hemorrhagic telangiectasia (HHT), a mutation (c.784A > G) in NAPG was found to co-segregate with the disease, and functional prediction showed it was deleterious and might change the conformational stability of the NAPG protein, potentially leading to HHT [1]. In the Chinese Han population, several SNPs in NAPG were associated with bipolar disorder, and certain haplotypes were also related to the disorder, supporting NAPG as a candidate for susceptibility to bipolar disorder [3]. In another study, NAPG was investigated as a potential common risk gene for bipolar disorder and schizophrenia in the Chinese population, though no abstract was available on its conclusion [2].
In fetal growth restriction, NAPG was among the top 5 mRNA genes with altered expression in newborn umbilical cord blood, maternal serum, and placental tissue [4]. In prostate cancer, SMR analyses revealed NAPG in prostate tissue was a potential drug target [5].
In conclusion, NAPG seems to be involved in multiple disease-related processes. Its mutations and polymorphisms may contribute to the development of hereditary hemorrhagic telangiectasia and bipolar disorder. The study of NAPG in these disease models helps to understand the molecular mechanisms underlying these diseases, providing potential insights for diagnosis, treatment, and prevention.
