PSMD2, encoding 26S proteasome non-ATPase regulatory subunit 2, is a key component of the ubiquitin-proteasome system, which is crucial for regulating intracellular protein levels. This system collaborates with the autophagy-lysosomal system in maintaining protein homeostasis, a process often dysregulated in malignancy [1].
PSMD2 has been shown to act as an oncogene in multiple cancers. In renal cell carcinoma treated with immune checkpoint and tyrosine kinase inhibitors, PSMD2 overexpression is linked to resistance, decreased progression-free survival, and impaired function of tumor-infiltrating lymphocytes [3]. In lung adenocarcinoma, high PSMD2 expression is correlated with poor prognosis and immune cell infiltration levels [4]. In nasopharyngeal carcinoma, DNAJA4 suppresses metastasis via PSMD2-mediated MYH9 degradation [5]. In breast cancer, RACK1 promotes cancer progression by competitively inhibiting β-catenin's binding to PSMD2, enhancing β-catenin stability [6]. Also in breast cancer, PSMD2 regulates cell proliferation and cell cycle progression by modulating p21 and p27 proteasomal degradation [7]. In HepG2 cells, PSMD2 promotes proliferation by facilitating cellular lipid droplet accumulation [8].
