Anxa10, a member of the annexin family, is a membrane-bound calcium-dependent phospholipid-binding protein. Its functions are still being elucidated, but it is likely involved in various biological processes and is associated with multiple pathways, with potential importance in understanding disease mechanisms. Mouse models with Anxa10-related genetic modifications are valuable research tools for exploring its functions [1,4].
Knockdown of Anxa10 in colorectal cancer cells reduces cell survival, proliferation, and migration, inhibits lung metastasis, and induces ferroptosis by inhibiting autophagy-mediated TFRC degradation [1]. In melanoma, Anxa10 knockout profoundly reduces cell migration and metastatic activity, and induces the N-to E-cadherin switch [2]. In hepatocellular carcinoma, upregulation of Anxa10 inhibits cell proliferation and migration [3]. In urothelial carcinoma, Anxa10 expression is inversely associated with tumor stage and grade [5]. In esophageal squamous cell carcinoma, Anxa10 induced by interaction with tumor-associated macrophages promotes cell growth [6]. In papillary thyroid carcinoma, knockdown of Anxa10 inhibits cell proliferation and promotes apoptosis by down-regulating TSG101 and inactivating the MAPK/ERK signaling pathway [7]. In bladder cancer, low Anxa10 expression is associated with shorter progression-free survival and faster cell growth [8].
In conclusion, Anxa10 plays diverse roles in multiple diseases, affecting processes such as cell survival, proliferation, migration, and metastasis. Mouse models, especially those with Anxa10 knockout or knockdown, have been crucial in revealing its functions in cancer-related biological processes, providing insights into potential therapeutic targets for various cancers.
