Fhl5, also known as four-and-a-half LIM (LIN-11, Isl-1, MEC-3) domain 5, is involved in multiple biological processes. It has been linked to the regulation of smooth muscle cell contraction, and is associated with pathways related to vascular remodeling. In addition, it may play a role in transcriptional regulation, as it can interact with CREM, a DNA-binding transcriptional regulator [3]. Fhl5 is also expressed in tissues like gill pillar cells, heart, and skeletal muscle, and may be related to maintaining cell integrity and dynamics in pillar cells [4].
In terms of disease associations, genome-wide association studies have implicated Fhl5 in vascular diseases such as coronary artery disease and myocardial infarction. In smooth muscle cells, FHL5 overexpression promotes vascular calcification and dysregulates extracellular matrix organization and calcium handling processes under pro-calcifying conditions [1]. Genetic polymorphisms of Fhl5 are associated with diabetes mellitus risk in the Chinese population [2].
