Mfge8, also known as Milk fat globule-epidermal growth factor 8, is a secreted protein that plays diverse roles in various biological processes. It binds to integrin families, such as αVβ3/αVβ5, α8β1, and integrin β3, and activates multiple signaling pathways including PI3K/AKT, ERK/AKT, and Src-FAK-STAT3. It is involved in processes like angiogenesis, fibrosis, efferocytosis, insulin signaling, neurogenesis, cancer progression, and gastric muscle contraction, highlighting its overall biological importance. Genetic models can be valuable in further elucidating its functions [1,2,3,4,5,6,7].
In esophageal squamous cell carcinoma (ESCC), the MFGE8 secreted by cancer-associated fibroblasts (CAFs) promotes tumor angiogenesis, proliferation, drug resistance, and metastasis by activating PI3K/AKT and ERK/AKT pathways via binding to Integrin αVβ3/αVβ5 receptors. Targeting CAFs-secreting MFGE8 or its receptor significantly inhibits ESCC growth and metastasis in vivo [1].
In colorectal cancer, CRC-derived extracellular vesicles containing MFGE8 enhance macrophage efferocytosis of cisplatin-induced apoptotic CRC cells by activating the macrophage αvβ3-Src-FAK-STAT3 signaling pathway. MFGE8 knockout CRC cell lines show that their EVs fail to upregulate macrophage efferocytosis in vitro and in vivo, suggesting MFGE8's role in promoting drug resistance [3].
In rats with experimental subarachnoid hemorrhage (SAH), exogenous recombinant human MFGE8 (rhMFGE8) administration promotes adult hippocampal neurogenesis and improves neurobehavioral recovery by activating the integrin β3/Akt pathway. Silencing integrin β3 or inhibiting the PI3K/Akt pathway abolishes the effects of rhMFGE8 [5].
In conclusion, Mfge8 has a wide-reaching impact on numerous biological processes. Gene knockout models, as seen in ESCC, colorectal cancer, and SAH studies, have revealed its role in cancer progression, drug resistance, and neurogenesis. These findings contribute to understanding the mechanisms of these diseases and suggest potential therapeutic targets related to Mfge8.
